David Sinclair on NMN: What He Says About NAD+, Benefits and Side Effects

NMN has become one of the most talked-about compounds in the healthy-ageing world, and few researchers are more closely associated with the public conversation around NAD+ biology than Professor David Sinclair.

In an interview with Auren Hoffman, Sinclair was asked directly why he is interested in NMN, whether he takes it himself, what the human evidence suggests, and what is known about possible side effects. It is a useful conversation because Sinclair is enthusiastic about the biology while also making an important distinction that is sometimes lost online: interest in NMN is not the same thing as proof that it extends human lifespan.

Why does David Sinclair talk about NMN?

Sinclair starts with NAD+, a molecule involved in essential cellular chemistry. He describes his laboratory's long-running interest in sirtuins — a family of proteins involved in cellular responses to stress — and explains that sirtuin activity depends on NAD+.

His argument is that NAD+ biology is relevant to ageing because NAD+ levels tend to change as we get older. NMN, or nicotinamide mononucleotide, matters in this context because it is a precursor the body can use to make NAD+.

In the interview, Sinclair says that human studies have shown NMN can raise NAD+ levels, describing increases of around two- to threefold in some research. That is an important distinction: raising a biological marker such as NAD+ is measurable, but it does not automatically prove a particular long-term health outcome.

Does David Sinclair take NMN himself?

Yes. Sinclair says in the interview that he takes NMN and had been doing so for approximately eight to ten years at the time of the conversation.

He is also unusually explicit about something worth noting: he says he does not sell NMN and that companies have incorrectly claimed an association with him. His personal use should therefore be understood as exactly that — a personal decision by a researcher interested in the field, not an endorsement of Little Ox or any other commercial NMN product.

Personal use by a scientist can be interesting context. It is not clinical evidence on its own.

What benefits does Sinclair discuss?

Sinclair refers to emerging human research around NAD+ precursors and mentions measures such as performance, endurance and six-minute walking tests. He presents this as an active area of human research rather than a finished scientific story.

That nuance matters. An improvement in a particular trial endpoint does not mean NMN has been proven to slow ageing generally, prevent disease or extend life. Different studies use different populations, doses, durations and endpoints, and the long-term evidence base remains much less developed than the basic biology.

Does David Sinclair say NMN extends lifespan?

No — and this is probably the most important line in the interview.

“It’s not proving that it extends lifespan.”

Sinclair discusses early animal work and says there was still much more to do. Whatever someone thinks about the potential of NAD+ biology, this is a useful guardrail: the interview does not provide evidence that taking NMN has been proven to extend human lifespan.

This is also why we avoid describing NMN as a miracle anti-ageing drug. The responsible position is to separate what has been observed in laboratory and short-term human research from the much bigger claims that would require robust long-term clinical evidence.

What does Sinclair say about NMN side effects?

Hoffman asks Sinclair directly whether taking NMN appears to have side effects or biological trade-offs.

Sinclair says he was not aware at that point of a tangible, proven risk emerging from the human subjects studied, and he says adverse events had not been seen in the human clinical work he was referring to. But he does not present the issue as closed.

He specifically mentions small animal studies involving unusual genetically inbred, immunodeficient mice that had raised questions about cancer spread in those highly specific models. His interpretation is that these findings had not stopped human clinical research, but he makes clear that he wants risks to be identified if they emerge.

That is a more useful takeaway than either extreme — neither “NMN is completely risk-free” nor “an animal finding proves NMN is dangerous” is supported by this interview. Safety evidence evolves as larger and longer human studies accumulate.

If you have a medical condition, take prescription medication, are pregnant or breastfeeding, or have a specific concern about supplementation, speak to an appropriately qualified healthcare professional before taking NMN or any new supplement.

One of Sinclair’s strongest points is actually about scientific scepticism

Later in the discussion, Sinclair makes a broader point that applies particularly well to longevity supplements: do not accept a claim simply because a podcaster, doctor or public figure repeats it. Go back to the underlying paper and look at what the data actually show.

Sinclair also stresses that lifestyle matters. Exercise, diet, fasting and other everyday behaviours feature heavily in his discussion of healthy ageing. Supplements should not be treated as a replacement for the fundamentals of health.

So, what should a UK buyer take from the interview?

There are five useful conclusions:

  • NMN is an NAD+ precursor. That is the biological reason researchers are interested in it.
  • Sinclair says he personally takes NMN. That is personal context, not proof of efficacy and not a product endorsement.
  • Human research can measure changes in NAD+ and other endpoints. Those findings should not be inflated into claims the studies were not designed to prove.
  • Human lifespan extension is not proven. Sinclair says this directly.
  • Quality and evidence matter. If you choose to take NMN, look beyond hype and examine dose, formulation, testing and transparency.
