NMN vs NR: Which NAD+ Precursor Should You Actually Take?
If you've been researching NAD+ supplements, you've almost certainly encountered two competing options: NMN (Nicotinamide Mononucleotide) and NR (Nicotinamide Riboside). Both are marketed as ways to raise NAD+ levels. Both have genuine scientific support. So which should you choose?
The honest answer is that the science slightly favours NMN — but the full picture is more nuanced than most supplement brands will tell you.
What Are NMN and NR?
Both NMN and NR are molecules that your body uses to produce NAD+. They're related — NR is actually a precursor to NMN. The metabolic pathway looks like this:
NR → NMN → NAD+
This means NMN is one step closer to NAD+ than NR. When you take NR, your body has to convert it to NMN first before it can be used to produce NAD+. When you take NMN directly, you skip that step.
The Absorption Question
For years, a key argument in favour of NR was that NMN couldn't be absorbed intact — that it had to be broken down to NR in the gut before entering cells. This was used to suggest NR was effectively just as direct a precursor as NMN.
That argument has been significantly undermined by a 2019 study identifying a specific transporter protein (Slc12a8) that allows NMN to be directly absorbed into intestinal cells without conversion to NR first. Further research has shown that NMN appears in the bloodstream within minutes of oral supplementation.
In short: NMN can be absorbed directly, and it raises NAD+ levels effectively.
Clinical Evidence: What the Trials Show
Both NMN and NR have been studied in human trials, and both have demonstrated the ability to raise blood NAD+ levels. Key differences:
- Dose efficiency: NMN appears to raise NAD+ levels at comparable or lower doses than NR
- Bioavailability: Current evidence suggests NMN may have slightly superior bioavailability, particularly in older adults
- Research depth: NR has a larger body of human trial data simply because it has been studied for longer. NMN research is catching up rapidly
- Cognitive effects: Some NR studies have specifically examined cognitive outcomes. NMN studies have focused more on energy, muscle function and metabolic health
Cost Comparison
NR is generally more expensive than NMN per milligram of active ingredient. This is partly historical — NR had early patent protection that kept prices high. As NMN manufacturing has scaled up, the price gap has narrowed, but NMN typically remains better value for money at equivalent doses.
What About NMN Plus with Resveratrol?
One area where NMN genuinely has an edge is in combination formulas. NMN works synergistically with Trans-Resveratrol — a sirtuin activator found in red grapes and dark berries. NAD+ activates sirtuins, and resveratrol amplifies that activation. This combination has been popularised by researchers like Dr David Sinclair at Harvard.
There is no equivalent well-researched NR-plus-resveratrol combination with the same level of scientific interest.
The Verdict
If you're choosing between NMN and NR purely on current evidence, NMN has a slight edge — it's one step closer to NAD+, appears to be efficiently absorbed, and offers the option of the synergistic NMN + Resveratrol combination. NR remains a valid option, particularly if you've already been taking it with good results.
What matters most is consistency. Neither NMN nor NR works if you take it for two weeks and stop.
