NMN for Women: What the Evidence Says About Menopause, Energy and Healthy Ageing
NMN is often discussed in the context of general longevity science — but there are specific reasons why women, particularly those in their 40s and 50s, may benefit most from NAD+ restoration.
As a GP who sees perimenopausal and postmenopausal women every week, I want to share what the clinical evidence shows — and be honest about where we have strong data and where the research is still emerging.
The Connection Between NAD+ and Female Hormonal Health
NAD+ decline and oestrogen decline often happen simultaneously — both accelerating in the 40s and reaching significant levels by the time of menopause. This isn't coincidental. Oestrogen plays a role in regulating several NAD+ biosynthetic enzymes, meaning that as oestrogen falls, the body's ability to produce NAD+ is further compromised beyond simple age-related decline.
The result is a compounding effect: women in perimenopause and menopause experience the standard age-related NAD+ decline AND the additional hormonal disruption to NAD+ production. This makes NAD+ restoration particularly relevant for this group.
What the Clinical Evidence Shows for Women
A 2021 clinical trial published in Science found that NMN supplementation significantly improved muscle insulin sensitivity in postmenopausal women with prediabetes — one of the clearest demonstrations of a clinically meaningful benefit in a specific female population. The women in this trial showed improvements in skeletal muscle insulin sensitivity comparable to what you might expect from a meaningful exercise intervention.
Other research has shown that NAD+ supports mitochondrial function in muscle tissue — relevant to the muscle loss (sarcopenia) that accelerates after menopause. Higher NAD+ levels support the cellular machinery that maintains muscle quality and recovery.
NMN and Energy During Perimenopause
Fatigue is the most commonly reported symptom in perimenopause — ahead of hot flushes, sleep disturbance and mood changes in many surveys. Much of this fatigue is directly cellular: declining mitochondrial function reduces the ATP output of individual cells, creating a systemic energy deficit that no amount of coffee addresses.
NMN supports mitochondrial function by restoring the NAD+ that the electron transport chain requires. This is not the same as a stimulant effect — it's addressing a root cause of cellular metabolism deficit. The effect builds over weeks, not hours.
NMN, Sleep and Circadian Rhythm
Sleep disruption is near-universal in perimenopause. NAD+ is closely involved in regulating circadian clock genes — the molecular machinery that governs sleep-wake cycles. Lower NAD+ levels are associated with disrupted circadian signalling, which may compound the sleep disruption from hormonal changes.
Many women taking NMN report improved recovery and overnight routine as one of the first noticeable changes, typically within 2–4 weeks of consistent supplementation. The mechanism — circadian NAD+ support — is biologically plausible, though large-scale trials specifically measuring sleep outcomes in perimenopausal women are still ongoing.
NMN and Skin Health
NAD+ is essential for sirtuin activation — particularly SIRT1 and SIRT3, which regulate cellular repair processes including those that maintain skin integrity and collagen synthesis. As NAD+ declines, the cellular repair mechanisms that keep skin elastic and healthy become less efficient.

NMN should not be taken during pregnancy or breastfeeding — not because of known harm, but because there is insufficient data on safety in these specific populations.
